Description | Mouse anti Human Fibroblasts/Epithelial Cells antibody, clone D7-FIB recognizes a ~112kDa molecule expressed on the cell surface of human fibroblasts. The antigen is not expressed by peripheral blood cells, and is found at low levels on a minority of melanoma cell lines. Studies upon the antigen have shown it to be sensitive to SDS, but resistant to trypsin, tunicamycin and collagenase.
D7-FIB has also been found to bind to epithelium, myoepthelium, smooth muscle and some leucocytes. D7-FIB is useful as a cell membrane marker to characterize chondrocyte differentiation giving a positive reaction with dedifferentiated human chondrocytes, and negative with differentiated chondrocytes (van Osch et al. 2001). |

Specificity | NB600-777 recognizes a 112kD molecule expressed on the cell surface of human fibroblasts and epithelial cells. In peripheral blood the antibody stains myeloid cells and a very small number of lymphocytes. Studies upon the antigen have shown it to be sensitive to SDS, but resistant to trypsin, tunicamycin and collagenase. In immunohistological studies the antibody has also been found to bind to epithelium, myoepthelium, smooth muscle and some leucocytes. D7-FIB has been shown to be useful as a cell membrane marker to characterize chondrocyte differentiation giving a positive reaction with dedifferentiated human chondrocytes, and negative with differentiated chondrocytes (3). This product is routinely tested in flow cytometry on the KG1 cell line. |

Application Notes | Flow Cytometry: Use 10 ul of the suggested working dilution to label 10^6 cells in 100 ul. The epitope recognised by this antibody is reported to be sensitive to formaldehyde fixation and tissue processing. The use of acetone fixation for frozen sections is recommended. This antibody does not work in Immunohistochemistry-Paraffin. Use in Immunocytochemistry/immunofluorescence reported in scientific literature (PMID 27416040). |
