Background

The gamma globin genes (HBG1 and HBG2) are normally expressed in the fetal liver, spleen and bone marrow. Two gamma chains together with two alpha chains constitute fetal hemoglobin (HbF) which is normally replaced by adult hemoglobin (HbA) at birth. In some beta-thalassemias and related conditions, gamma chain production continues into adulthood. The two types of gamma chains differ at residue 136 where glycine is found in the G-gamma product (HBG2) and alanine is found in the A-gamma product (HBG1). The former is predominant at birth. The order of the genes in the beta-globin cluster is: 5'-epsilon -- gamma-G -- gamma-A -- delta -- beta--3'.

  1. May I know if NB110-41084 is specific only to the gamma chain of Hemoglobin F and does not exhibit cross-reactivity with the Hemoglobin A chain? There are several anti-hemoglobin F antibodies on the market that exhibit cross-reactivity with the A chain; we are looking for an antibody that does NOT exhibit residual activity with the A chain.
    • For NB110-41084 please see Summary section of the datasheet. Specificity: "Hemoglobin F. No antibody was detected against hemoglobin A1, A2, S or human serum proteins." It appears that from testing, this antibody does not cross-react with Hemoglobin A1 or A2.
  2. Hello! I am looking at product NB110-41084, Fetal Hemoglobin antibody. Can you tell me if this cross reacts with Regular Hemoglobin typically found in adults?
    • No reactivity was detected against hemoglobin A1, A2, S or human serum proteins. So no, it will not crossreact with adult hemoglobin
