LDL Receptor: Low Density, High Importance

Fri, 01/03/2014 - 13:09

The low density lipoprotein receptor coordinates the metabolism of cholesterol, an essential component of the mammalian cell plasma membranes. Study of this carefully balanced system has led to an enhanced understanding of cholesterol homeostasis at the cellular level. Receptor-mediated endocytosis (RME) is an important mechanism of metabolic regulation. Garbarino’s group used LDL receptor antibody and small hairpin RNA knockdown to show that knockdown of STARD4 in hepatocytes disrupts cholesterol trafficking between the plasma membrane, endoplasmic reticulum (ER), and endocytic recycling compartment (ERC) in a nonvesicular transport mechanism (1).

The novel bioactive compound evodiamine was found to confer a transient receptor potenitial vanilloid type 1 (TRPV1) dependent protection against atherosclerosis as well as a TRPV-1 independent action against obesity in studies with low density lipoprotein (LDL) receptor antibody (2). Infertility studies using low density lipoprotein (LDL) receptor antibody found that the scavenger receptor class B type 1 (SR-BI) lipoprotein receptor decreases progesterone secretion in human granulose cells and may explain the lowered progesterone levels in certain women (3). Researchers have employed low density lipoprotein (LDL) receptor antibody to characterize the cellular uptake and degradation of beta amyloid peptide by astrocytes (4). They were able to show that the LDL receptor directly binds amyloid protein and enhances it degradation through lysosomal targeting. Other neural studies with low density lipoprotein (LDL) receptor antibody in showed for the first time that the MPTP compound activates the apoE cascade in mice striatum in Parkinson’s disease (5).
