Protein expression levels were assessed for phosphorylated or total forms of PERK, eIF2 alpha & for CHOP, BiP, XBP1-s & XBP1-u proteins. Efficiency of rapamycin to inhibit mTORC1 pathway was also checked by immunoblot with phosphorylated & total forms of p70S6K1 & 4E-BP1. alpha -tubulin was used as internal control. (B & C) Cells were incubated with 10nM rapamycin for 1 h (B) or 24 h (C). Immunoblots for phosphorylated & total forms of PERK, eIF2 alpha , p70S6K1 & 4E-BP1 or for ATF4, CHOP, BiP, XBP1-s & XBP1-u protein expression were performed. alpha -tubulin was used as internal control. EVs isolated from TIS & control cells were immunoblotted to validate the protein expression of annexin V, Na/K ATPase & alpha -tubulin from the MS data. CD63 & HSP70 were used as loading controls. ARPE-19 cells were cultured up to 4 weeks on transwell inserts covered with laminin. The immunofluorescence image in (A) shows actin localization using rhodamine phalloidin. The cells are flat with stress fibers & very little circumferential actin microfilament bundles. The expression of CD147, a RPE marker, was detected using a specific antibody in the apical membrane domain (B). The expression of Na+, K+-ATPase using anti-alpha 1(C) & anti-beta 1 antibodies (D) was observed mostly at the basolateral membrane. Photomicrographs show immunofluorescence double-labeling for PAR1 with Sox2-positive (A), or PAR1 with Nestin-positive (C) multipotent neural stem cells (NSCs) within the lateral wall of the lateral ventricle (LV). RNAscope was used to identify cells expressing both PAR1 & Sox2 (B), or Nestin (D) RNA in NSCs of the adult SVZ. Arrow indicates an example of a double-labeled cell in each case, with arrowhead indicating a singly labeled cell (Scale bar = 10 μm).
