NPHP3-promoter plasmids were prepared from wildtype (wt.) promoter. (b–e) HeLa cells were transfected with pCMV-2B or pCMV-T beta 4 for 24 h. (b) HeLa cells were co-transfected with wildtype or mutant pEZX-PG02-NPHP3-promoter Gluc plasmid. & Gluc activity in cultured media was measured with luminometer using Gluc substrate. Bar graph indicates the mean of NPHP3-promoter activity. (c) Expression level of T beta 4 & NPHP3 transcripts were measured by RT-PCR.
