Epilepsy, Reflex: Disease Bioinformatics

Epilepsy is defined as a diverse set of neurological disorders characterized by seizures. These seizures occur due to abnormal electrical activity in the brain. Factors that may lead to epilepsy include, but are not limited to, brain trauma, gene mutations, strokes, brain cancer, and extensive drug and alcohol use. Seizures occur due to a large release of glutamate, which causes a spread of excitation throughout the brain, which propagates the electrical signal and can lead to neuronal death. Reflex epilepsy is a condition in which seizures can be provoked by an external stimulus or, in some cases, internal mental processes. Patients with reflex epilepsy may have seizures solely in response to specific stimuli and not suffer spontaneous seizures; however, a combination of the two is possible. About 50 million people have epilepsy, with 80% of that population located in developing countries. It is possible to control epilepsy with medicine, but many cases may require surgery in order to relieve symptoms.

Western Blot: iNOS Antibody [NB300-605] - Analysis of iNOS was performed by loading 20 ug of RAW264 whole cell lysate untreated (left lane) or stimulated with LPS at 1 ug/mL for 16 hours (right lane) and 10 uL of PageRuler Plus Prestained Protein Ladder onto a 4-20% Tris-Glycine polyacrylamide gel. Proteins were transferred to a nitrocellulose membrane and blocked with 5% Milk in TBST for at least 1 hour. The membrane was probed with an iNOS Rabbit polyclonal antibody at a dilution of 1:1000 overnight at 4C on a rocking platform, washed in TBST, and probed with a Goat anti-Rabbit IgG (H+L) Secondary Antibody, HRP conjugate at a dilution of 1:1000 for 1 hour. Immunocytochemistry/Immunofluorescence: Aurora A Antibody [NBP1-51843] - HeLa cells were fixed and permeabilized for 10 minutes using -20C MeOH. The cells were incubated with anti- (NBP1-51843) at 2 ug/ml overnight at 4C and detected with an anti-rabbit Dylight 488 (Green) at a 1:1000 dilution for 60 minutes. Alpha tubulin (DM1A) NB100-690 was used as a co-stain at a 1:1000 dilution overnight at 4C and detected with an anti-mouse Dylight 550 (Red) at a 1:1000 dilution for 60 minutes. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 100X objective and digitally deconvolved.Simple Western: Aurora A Antibody [NBP1-51843] - Simple Western lane view shows a specific band for Aurora A in 0.5 mg/ml of HeLa lysate.  This experiment was performed under reducing conditions using the 12-230kDa separation system.

Knockout Validated: MUC1 Antibody - BSA Free [NBP1-60046] - MUC1 Antibody [NBP1-60046] - C57BL/6 Muc1 KO mice and WT mice were undergoing intranasal instillation of 75 ug of lipopolysaccharide (LPS) at day 1. Between days 2 and 3, animals were exposed to cigarette smoke (or control air) of 6 cigarettes followed by 8 cigarettes (or control air) at days 4 and 5, and 10 cigarettes (or control air) at day 6. Vehicle or dexamethasone at 3 mg/kg/day and 10 mg/kg/day was administered orally once a day between day 1 and 6. Animals were sacrificed at day 6 and lungs were homogenized to analyze protein expression of Muc1 cytoplasmic tail (CT). Protein expression was normalized to B-actin.
