Beclin 1: Regulator of Autophagy and Apoptosis

Beclin 1 is the mammalian orthologue of the yeast Apg6/Vps30 gene. Beclin 1 can complement the defect in autophagy present in apg6 yeast strains and stimulate autophagy when overexpressed in mammalian cells (1) and can bind to Bcl2, an important regulator of apoptosis (2) suggesting a role in two fundamentally important cellular pathways: autophagy and apoptosis. Beclin 1 is mono allelically deleted in human breast and ovarian cancers and is expressed at reduced levels in tumors, and is present in a complex bound to glutamate receptor δ2 (GluRδ2) in the nervous system leading to the activation of autophagy and death of cerebellar Purkinje cells (3).

Studies using embryonic stem cells and targeted deletion of Beclin 1 in mice and subsequent detection by Beclin 1 antibodies suggest that Beclin 1 and autophagy are critical for maintenance of tissue homeostasis in vivo (4). Beclin1 has also been shown to control the protein stabilities of USP10 and USP13 by regulating their deubiquitinating activities. Since USP10 mediates the deubiquitination of p53, regulating deubiquitination activity of USP10 and USP13 by Beclin1 provides a mechanism by which Beclin1 controls the levels of p53 (5). Beclin 1 dysfunction has been implicated in many disorders, including cancer and neurodegeneration the organization and function of the Beclin 1 network in cellular homeostasis focusing on the cross-regulation between apoptosis and autophagy is an active area of research.
