The antibodies do not distinguish between isoforms. ROBO1 was expressed at a low level on precrossing axons & was highly upregulated on postcrossing axons in both WT & Nova1/2 dKO embryos. ROBO1-expressing axons displayed the same lateral positioning defect as anti-L1 labeled axons (compare with Figure 4B). Reducing Robo1(e6b+) partially rescued the lateral positioning defect in Nova1/2 dKO embryos, while reducing Robo2(e6b+) alone did not rescue.
