Mouse NKp46/NCR1 Antigen Affinity-purified Polyclonal Antibody induces IFN-gamma secretion in mouse natural killer (NK) cells activated with 25 ng/mL Recombinant Mouse IL-2 (Catalog # 402-ML) and 25 ng/mL Recombinant Mouse IL-12 (Catalog # 419-ML), in a dose-dependent manner, as measured using the Quantikine Mouse IFN-gamma ELISA Kit (Catalog # MIF00). The ED50 for this effect is typically0.4-2.4 μg/mL.

NKp46/NCR1 was detected in perfusion fixed frozen sections of mouse spleen using Goat Anti-Mouse NKp46/NCR1 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF2225) at 3 µg/mL for 1 hour at room temperature followed by incubation with the Anti-Goat IgG VisUCyte™ HRP Polymer Antibody (Catalog # VC004). NKp46/NCR1 was detected in immersion fixed paraffin-embedded sections of human tonsil using Mouse Anti-Human NKp46/NCR1 Monoclonal Antibody (Catalog # MAB1850) at 5 µg/mL for 1 hour at room temperature followed by incubation with the Anti-Mouse IgG VisUCyte™ HRP Polymer Antibody (Catalog # VC001). Before incubation with the primary antibody, tissue was subjected to heat-induced epitope retrieval using Antigen Retrieval Reagent-Basic (Catalog # CTS013). Tissue was stained using DAB (brown) and counterstained with hematoxylin (blue). Specific staining was localized to natural killer cellsin germinal center. Human peripheral blood mononuclear cells (PBMCs) were stained with (A) Mouse Anti-Human NKp46/NCR1 Monoclonal Antibody (Catalog # MAB1850) or (B) Mouse IgG2B Isotype Control (MAB0041) followed by Goat anti-Mouse IgG APC-conjugated Secondary Antibody (F0101B) and Rabbit Anti-Human NCAM-1/CD56 PE-conjugated Monoclonal Antibody (FAB24086P). Staining was performed using our Staining Membrane-associated Proteins protocol.

Formalin-fixed paraffin-embedded tissue sections of human tonsil were probed for NKp46 mRNA (ACD RNAScope Probe, catalog #312658; Fast Red chromogen, ACD catalog # 322750). Adjacent tissue section was processed for immunohistochemistry using mouse anti-human NKp46 monoclonal antibody (R&D Systems catalog # MAB1850) at 15ug/mL with overnight incubation at 4 degrees Celsius followed by incubation with anti-mouse IgG VisUCyte HRP Polymer Antibody (Catalog # VC001) and DAB chromogen (yellow-brown). Tissue was counterstained with hematoxylin (blue). Specific staining was localized to cell surface.
