Simple Western lane view shows a specific band for Caspase 1 in 1.0 mg/ml of HeLa lysate.  This experiment was performed under reducing conditions using the 12-230 kDa separation system.Activity of Casp1 in OHT-injured and normotensive control eyes. (A) Casp1 was detected by intraocular injection FLICA660-labeled substrate (green) in vivo 24 h after injury. Bright labeling (arrows) is evident in cells in the GCL and inner nuclear layer (INL) layers of the OHT-challenged retinas, a diffuse labeling of cell processes located in the IPL. Casp1 activity is diminished in Panx1-/- (Px1-/- OHT) retinas and WT retinas treated with probenecid (WT/Pbcd) at 12 h postinjury. Western Blot: Desmoglein-3 Antibody (5H10) [NBP1-78984] - Colocalisation of Dsg3 with E-cadherin-catenin complex. Co-IPs demonstrated that both endogenous and exogenous Dsg3 interacted with the E-cadherin-catenin complex in A431 stable lines (the empty vector control (V) and hDsg3.myc (D3)) as well as in HaCaTs. When pulled down with antibodies against the myc tag or with a mouse Dsg3 Ab 5H10 and blotted for E-cadherin and beta-catenin, both proteins were detected in D3 and, to a lesser extent, in V. Conversely, when pulled down with E-cadherin Ab HECD-1 and blotted for Dsg3 or the myc tag and Pg, these proteins were detected in both V and D3 cells. Note that this result also was confirmed in HaCaTs (HaC) where Dsg3 was expressed endogenously (far right blot). Dual RNAscope ISH-IHC: TLR4 Antibody (76B357.1) [NB100-56566] - FFPE tissue sections of human tonsil were probed for TLR4 mRNA (ACD RNAScope Probe, ACD catalog # 311281; Fast Red chromogen, ACD catalog # 322750). Adjacent tissue section was processed for immunohistochemistry using Mouse Monoclonal (Novus Biologicals catalog # NB100-56566) at 5ug/mL with 1 hour incubation at room temperature followed by incubation with anti-mouse IgG VisUCyte HRP Polymer Antibody (Catalog # VC001) and DAB chromogen (yellow-brown). Tissue was counterstained with hematoxylin (blue). Specific staining was localized to lymphocytes.Flow Cytometry: TLR4 Antibody (76B357.1) [NB100-56566] - Analysis using the Alexa Fluor (R) 647 conjugate of NBP2-27149. TLR4 expression on monocytes from human peripheral blood. PBMC were stained in a 2 color flow test, with CD14 PE version of this antibody and 1 ug of either isotype control (left) or TLR4-Alexa Fluor 647 (right). PPI negative, CD14+ cells were gated for analysis.
