Histones are nuclear proteins that are responsible for the nucleosome structure of the chromosomal fiber as they play a critical role in transcription regulation, DNA repair, DNA replication, and chromosomal stability. The HIST3H3 gene is intronless and encodes a 136 amino acid long, 15 kDA histone H3.1t protein that lacks polyA tails and obtain a palindromic termination element. The HIST3H3 gene is located separately from the other H3 genes on the cluster on chromosome 6p22-p21.3. HIST3H3 participates in PKA signaling, development notch signaling pathway, histone modification, and interferon pathway through interactions with genes HIST1H4B, HIST4H4, HIST1H4A, HIST1H4C, and HIST1H4D. HIST3H3 has been researched regarding its role in immunodeficiency, leukemia, prostatitis, huntington's disease, lupus, multiple sclerosis, laryngeal cancer, and lymphomas.
