Myc-tag: The "Monkey Wrench" of Proteomic Tools

c-Myc is a well-characterized transcription factor encoded by the c-Myc gene on human chromosome 8q24. This cellular proto-oncogene, also known as p62, is commonly activated in a variety of tumor cells and plays a crucial role in cellular proliferation, differentiation, apoptosis, and cell cycle progression. The myc-tag antibody contains a short sequence epitope that is widely and commonly used as a protein fusion tag. It is a very powerful investigative research toolfor applications such as immunochemistry, protein purification, ELISA purification, and protein localization (1).

Immunocytochemistry/Immunofluorescence: c-Myc Antibody

Hubbi, et. al. used the myc-tag antibody to demonstrate that MCM protein subunits from DNA helicase down-regulate proliferation through the hypoxia-inducible factor 1 pathway in response to hypoxia (2). Using myc-tag antibody immunoprecipitation studies, researchers in Kim’s lab were able to show that the signaling protein Notch1 complexes with its partner Jagged1 through proteasomal degredation of the Notch1 (3). Visualization studies with myc-tag antibody were able to examine the intracellular interactions, cell cycle distributions, and protein turnover rates of protein phosphatase 1 isoforms (4). The myc-tag antibody was used to determine the localization and interaction of FXYD3 complexes within the plasma membrane of colorectal cancer cells (5).
