Alternate Names for pCL-Ampho Retrovirus Packaging Vector
Background
The pCL-Ampho packaging vector is a part of the RetroMax expression system and has been designed to maximize recombinant-retrovirus titers in a simple, efficient, and flexible experimental system. By introducing a retroviral vector into a cell expressing retroviral proteins, retroviral particles (virions) are shed into the culture medium at the rate of 1 infectious particle/cell/day. Retrovirus tropism is determined at three levels. The first is simply a function of viral envelope protein, gp70. The envelope determines which cells the virus will enter. gp70 comes in three different flavors for gene therapy. The plasmid DNA can be amplified by transforming bacteria and growing them on LB/ampicillin (100 ug/ml) media.

I am having a retroviral system in place which allows me to infect mouse cell lines. The vectors I have are MIG.R (with my gene of interest), pCL-ECO and VSV-g. I know that my pCL-ECO vector is only suitable for infecting mouse cells. My question is: Is it possible to use my MIG.R and VSV-g vectors, and exchange the mouse specific pCL-ECO with pCL-Ampho or pCL-10A1 packaging vector which you both offer for purchase (Cat Numbers are 10045P, 10046P, 10047P). My aim is to infect human cell lines with my gene of interest.
- These vectors, while also sold separately, are components of the RetroMax system 10040K. The vectors are designed to be used as described in the 10040K manual. Yes, you are correct, pCL-ECO is for mouse and rat, and not for human. We have not evaluated the Retromax system with the MIG.R and VSV-g vectors, and can not promise they would be compatible with your vectors. The Retromax packaging vectors are designed to be used with the expression vectors in the Retromax Kit. However, the packaging vectors are widely published and you may be able to find some information in the literature relevant to your model system (search scholar.google.com and keywords with Retromax and vector name, or keys words with packaging vector name and expression vector name ).

I was curious if you had a map for the pcl-ampho vector?
- Unfortunately, we do not have a pCL-Ampho Vector Map in our records. This product was originally developed by Imgenex, a company we acquired at the beginning of last year, and it does not appear that the vector maps were supplied to us. We do have several publications referencing this product and you may find some of those publications useful: click on Reviews & Publications tab at NBP2-29541.

NBP2-29541 what is the sequence?
- As stated under "Notes," the sequence of NBP2-29541 is not available.You can find out more information about this plasmid in the manual, including a plasmid map on page 22. The map is for pCl-ECO but the only difference between the Eco and Ampho plasmids is the envelope gene.
