Goat anti-Mouse IgG1 Secondary Antibody Summary

Description
The lyophilized IgG (7S) fraction is shipped at ambient temperature and may be stored at 4C; prolonged storage at or below -20C. Prior to use, an aliquot is thawed slowly at ambient temperature, spun down again and used to prepare working dilutions by adding sterile phosphate buffered saline (PBS, pH 7.2). Repeated thawing and freezing should be avoided. Working dilutions should be stored at 4C, not refrozen, and preferably used the same day. If a slight precipitation occurs upon storage, this should be removed by centrifugation. It will not affect the performance of the product.
Immunogen
Pools of purified homogenous IgG1 isolated from pooled mouse serum. Freund's complete adjuvant is used in the first step of the immunization procedure.
Specificity
The reactivity of the antiserum is directed to the subclass IgG1 as tested against mouse sera and a panel of purified homogenous immunoglobulins. It does not react with other subclasses of IgG, IgG/Fab fragments, IgM and IgA or any non-Ig protein in mouse serum, as tested by immunoelectrophoresis and double radial immunodiffusion.

Reactivity Notes

This IgG fraction is not species-specific since inter-species cross-reactivity is a normal feature of antisera to immunoglobulins. However this product has been passed over appropriate immunoadsorbents to remove antibodies cross-reacting with Human immunoglobulins. This renders it specific for use in test systems containing material of Human origin (e.g. Human tissue/Mouse monoclonal antibody to a Human tissue constituent/anti Mouse Ig isotype-specific immunoconjugate. If the IgG fraction is to be used in the presence of material originating from another species, prior screening for cross-reactivity is essential an additional adsorption of the reagent may required to ensure intra-assay specificity. In double radial immunodiffusion a reaction with Rat has been observed.

Background

IgG is a monomeric immunoglobulin composed of two heavy chains and two light chains. There are four subclasses of the IgG: IgG1, IgG2, IgG3 and IgG4. Each molecule has two antigen binding sites. IgG is the most abundant immunoglobulin as well as the only isotype that can pass through the placent,a thereby providing protection to the fetus in its first weeks of life before, its own immune system has developed. IgG can bind to several different kinds of pathogens, for example viruses, bacteria and fungi, and it protects the body against them by complement activation (the classic pathway), opsonization for phagocytosis and neutralization of their toxins.

CD16 - Find me on macrophages, neutrophils and NK cells
CD16 is a lymphocyte Fc gamma type III low-affinity receptor for IgG and is represented by two similar genes, CD16A (Fc gamma RIII A) and CD16B (Fc gamma RIIIB). CD16A exists as a heterooligomeric polypeptide-anchored form in macrophages and NK cells....  Read full blog post.
