Rabbit anti-Guinea Pig IgG1 Secondary Antibody Summary

Description
The lyophilized antiserum is shipped at ambient temperature and may be stored at 4C; prolonged storage at or below -20C. Dilutions may be prepared by adding phosphate buffered saline (PBS, pH 7.2). Repeated thawing and freezing should be avoided. If a slight precipitation occurs upon storage, this should be removed by centrifugation. It will not affect the performance of the antiserum. Diluted antiserum should be stored at 4C, not refrozen, and preferably used the same day.
Immunogen
Pools of purified normal IgG1 isolated from pooled guinea pig serum. Freund's complete adjuvant is used in the first step of the immunization procedure.
Specificity
The reactivity of the antiserum is directed to the Fc subunit of subclass IgG1. It does not react with other IgG2, IgG/Fab fragments, IgM and IgA or any non-Ig protein in guinea pig serum, as tested by immunoelectrophoresis and double radial immunodiffusion.

Application Notes
Precipitation assays. In immunoelectrophoresis use 2 ul or equivalent against 120 ul antiserum. In double radial immunodiffusion (Ouchterlony) use a rosette arrangement with 10 ul antiserum in a 3 mm diameter center well and 2 ul serum samples (neat and diluted) in 2 mm diameter peripheral wells.

Reactivity Notes

This IgG fraction is not species-specific since inter-species cross-reactivity is a normal feature of antisera to immunoglobulins. Cross-reactivity of this product has not been tested in detail.

Background

IgG is a monomeric immunoglobulin composed of two heavy chains and two light chains. There are four subclasses of the IgG: IgG1, IgG2, IgG3 and IgG4. Each molecule has two antigen binding sites. IgG is the most abundant immunoglobulin as well as the only isotype that can pass through the placent,a thereby providing protection to the fetus in its first weeks of life before, its own immune system has developed. IgG can bind to several different kinds of pathogens, for example viruses, bacteria and fungi, and it protects the body against them by complement activation (the classic pathway), opsonization for phagocytosis and neutralization of their toxins.

CD16 - Find me on macrophages, neutrophils and NK cells
CD16 is a lymphocyte Fc gamma type III low-affinity receptor for IgG and is represented by two similar genes, CD16A (Fc gamma RIII A) and CD16B (Fc gamma RIIIB). CD16A exists as a heterooligomeric polypeptide-anchored form in macrophages and NK cells....  Read full blog post.
