Western blot shows lysates of mouse liver tissue. Gels were loaded with 12 µg, 6.5 µg, and 3 µg of tissue lysate. PVDF membrane was probed with 1 µg/mL of Goat Anti-Mouse MMR/CD206 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF2535) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF019). A specific band was detected for MMR/CD206 at approximately 180 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

MMR/CD206 was detected in perfusion fixed frozen sections of mouse testis using Goat Anti-Mouse MMR/CD206 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF2535) at 5 µg/mL overnight at 4 °C. Tissue was stained using the Anti-Goat HRP-DAB Cell & Tissue Staining Kit (brown; Catalog # CTS008) and counterstained with hematoxylin (blue). M1 and M2 phenotype in spinal cord after intraplantar IL-1 beta . Wild-type (WT) and LysM-G protein–coupled receptor kinase (GRK)2+/− mice received an intraplantar injection of 1 ng IL-1 beta . At 15 hours after injection, spinal cord was collected, and frozen sections of (A) lumbar spinal cord (L2 to L5) and as control (B) thoracic spinal cord (T6 to T10) were stained for M1 (CD16/32) and M2 (CD206 and arginase-I) phenotypic markers. PVDF Membrane was probed with 1 µg/mL of Goat Anti-Human MMR/CD206 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF2534) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF019). A specific band was detected for MMR/CD206 at approximately 185 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 8.

MMR/CD206 was detected in immersion fixed paraffin-embedded sections of human liver using Goat Anti-Human MMR/CD206 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF2534) at 3 µg/mL for 1 hour at room temperature followed by incubation with the Anti-Goat IgG VisUCyte™ HRP Polymer Antibody (VC004). Before incubation with the primary antibody, tissue was subjected to heat-induced epitope retrieval using Antigen Retrieval Reagent-Basic (CTS013). Tissue was stained using DAB (brown) and counterstained with hematoxylin (blue). Specific staining was localized to sinusoids. Staining was performed using our protocol for IHC Staining with VisUCyte HRP Polymer Detection Reagents.

Simple Western lane view shows lysates of human immature dendritic cells, loaded at 0.2 mg/mL. A specific band was detected for MMR/CD206 at approximately 201 kDa (as indicated) using 10 µg/mL of Goat Anti-Human MMR/CD206 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF2534) . This experiment was conducted under reducing conditions and using the 12-230 kDa separation system.

Cells were fixed with 4% PFA and then permeabilized with 0.1% saponin. Immunocytochemistry/Immunofluorescence: CD68/SR-D1 Antibody (KP1) [NB100-683] - A431 cells were fixed in 4% paraformaldehyde for 10 minutes and permeabilized in 0.05% Triton X-100 in PBS for 5 minutes. Western Blot: CD68/SR-D1 Antibody (KP1) [NB100-683] - Total protein from mouse Raw cells was separated on a 7.5% gel by SDS-PAGE, transferred to PVDF membrane and blocked in 5% non-fat milk in TBST. The membrane was probed with 2.0 ug/ml anti-CD68 (NB100-683) in blocking buffer and detected with an anti-mouse HRP secondary antibody using NovaLume chemiluminescence detection reagent (NPB2-61915).

Flow Cytometry: CD68/SR-D1 Antibody (KP1) [NB100-683] - An intracellular stain was performed on A431 cells with CD68/SR-D1 [KP1] Antibody NB100-683AF647 (blue) and a matched isotype control (orange). Dual RNAscope ISH-IHC: CD68/SR-D1 Antibody (KP1) [NB100-683] - NOS2 mRNA (red) and CD68 protein (green) were detected in formalin-fixed paraffin-embedded tissue sections of human breast cancer. ACD's Integrated Co-Detection Workflow was performed using ACD RNAScope Probe Hs-NOS2 and CD68/SR-D1 antibody (KP1) at 1:100 dilution. Tissue was stained on Leica Bond RX using RNAscope (TM) 2.5 LS Reagent Kit-RED, BOND Polymer Refine Detection (DAB) and Hematoxylin, BOND Polymer Refine Red Detection and Hematoxylin and RNAscope (TM) 2.5 LS Green Accessory Pack. Tissue was counterstained with 50% hematoxylin (blue).

CD68/SR-D1 (KP1) was detected in immersion fixed A431 human skin carcinoma cell line using Mouse anti-CD68/SR-D1 (KP1) Protein-G purified Monoclonal Antibody conjugated to DyLight 550 (Catalog # NB100-683R) (red) at 10 µg/mL overnight at 4C. Cells were counterstained with DAPI (blue). Cells were imaged using a 100X objective and digitally deconvolved.

An intracellular stain was performed on A431 human skin carcinoma cell line using Mouse anti- CD68/SR-D1 (KP1) Protein-G purified Monoclonal Antibody conjugated to DyLight 550 (Catalog # NB100-683R, blue histogram) or matched control antibody (orange histogram) at 2.5 µg/mL for 30 minutes at RT.
