The affects of Perilipin 2 on diet and metabolism

Perilipin 2 belongs to the Perilipin family, which consists of proteins that coat intracellular lipid storage droplets. Perilipin 2 in particular is involved in lipid globule surface membrane composition, and has also been implicated in the development and maintenance of adipose tissue. Contrary to previous findings, Perilipin 2 is found in a variety of cells aside from adipocytes, ranging from fibroblasts to skin cells. Overall, perilipins regulate cellular lipid metabolism and homeostasis.  On the contrary, excess lipid stores are linked to both exercise and obese conditions.  Maintaining the correct balance of lipid storage is very important to avoid lipocytotoxicity and is the focus of obese and metabolism related pathologies. 

Perilipin 2 antibody

Perilipin-2/ADFP Antibody [NB110-40877] - Simple Western lane view shows a specific band for ADFP in 0.5 mg/ml of HepG2 lysate. This experiment was performed under reducing conditions using the 12-230 kDa separation system.

There are two notable pathologies associated with imbalances in lipid storage; obesity and fatty liver disease.  Nishikawa et al studied the effects of resveratrol on mice with fatty liver to investigate the role of liver macrophages with the use of a Perilipin 2 antibody.  Resveratrol is a phytoalexin polyphenolic compound found in grapes, berries, and peanuts. The focus of the potential rescue effect of resveratrol in livers with high lipid accumulation is centered on the need for prevention of lifestyle-related diseases.
In order to research effectiveness of diet and exercise on obesity, Vigelso used a Perilipin 2 antibody to discover an interesting correlation between Perilipin 2 and aerobic activity when they measured Perilipin 2 protein levels after activity performed by old and young men.  Specifically, older men experienced an increase in Perilipin 2 and intramuscular triglyceride (IMTG).  It was however, unclear which may have lead to the increase of the other.  However, their lab did conclude that age related metabolic inflexibility was present, due to body fat percentages and IMTG levels.   

Conversely, McManaman found that Perilipin 2 null mice were protected against diet induced obesity, adipose inflammation and fatty liver disease. They used a Perilipin 2 antibody in immunohistochemistry in adipose tissue from their Plin2-null mice to show that there was a loss of inflammatory foci and a limited degree of liver pathology after a 45% fat diet.  Their results had a clear finding of how important the Perilipin 2 antibody is to the homeostasis of fatty liver disease. All in all, Perilipin 2 is a dynamic protein that is both increased in high fat diets, but decreased in Perilipin 2-null animals despite increase in food intake.
