ABCG1 (ATP-binding cassette sub-family G member 1) is a transporter protein that is primarily involved in macrophage lipid homeostasis. It is a member of the superfamily of ATP-binding cassette (ABC) transporters and localizes to intracellular compartments associated with endoplasmic reticulum and golgi membranes. Its expression is highest in macrophage-rich tissue such as spleen, lung, thymus, and brain. ABCG1 is expressed on the cell surface and in intracellular compartments of cholesterol-laden macrophages.

The ABCG1 protein forms heterodimers with other heterologous ABCG partners. In conjunction with ABCA1, ABCG1 allows the export of a variety of lipids such as cholesterol and phospholipids. Moreover, ABCG1 plays role in T cells/inflammation and brain lipid homeostasis. In general, the entire ATP-binding cassette transporter gene family is involved in the regulation of the amount of dietary cholesterol retained in the body. ABCG1 antibodies are heavily used in atherosclerosis disease studies. Khan, et. al. used ABCG1 antibodies in their studies on the GGTase-I enzyme on RHO signaling pathways in mice (1). They were able to show that targeting GGTase-I eventually results in reduced atherosclerosis despite increased inflammation. Horie, at. al. used ABCG1 antibodies to assess the impact of a microRNA-33 genetic deficiency on progression of atherosclerotic plaques and were able to show that the deficiency released HDL-C, increased cholesterol efflux from macrophages, and prevented atherosclerotic progression (2). Studies with ABCG1 antibodies and cholesterol efflux have found interesting links to immunity and cardiprotection (3). The diverse use of these antibodies is demonstrated by a breast cancer study using ABCG1 antibodies to verify that conjugated linoleic acids (CLA) may have a future as anti-tumor agents in breast cancer (4).
