ABCA1 is a key gatekeeper influencing intracellular cholesterol transport, and is an important member of a multifamily of cAMP-dependent anion transporter cell membrane proteins that regulate reverse cholesterol efflux from cells in peripheral tissues to apolipoprotein A1. ABCA1 has a wide expression profile with highest expression levels found in macrophages. ABCA1 antibody helped determine the role of Caveolin 1 in cellular cholesterol homeostasis in caveolin-1 deficient knockout mice (1). University of Washington researchers used ABCA1 antibodies to validate ABCA1 as an anti-inflammatory receptor, helping to explain its protective role against cardiovascular disease (2).

Real-time PCR and immunoblotting with ABCA1 antibody alloawed Sporstol’s group to generate expression pattern profiles of cholesterol metabolism targets such as scavenger receptor type B class I (SR-BI), ABCA1, and ABC transporter G1 (ABCG1) (3). More detailed ABCA1 antibody studies on ABCA1 regulation have found that the oxysterol-binding protein (OSBP1) involved in sterol sensing and transport does negatively regulate cytoplasmic ABCA1 activity through protein destabilization (4). Zhou, et al. determined with the ABCA1 antibody that low density lipoprotein receptor (LDLR) deletions such as those found in atherosclerosis and familial hypercholesterolemia critically regulate cholesterol efflux and ABCA1 macrophage expression (5). They were not the only group to examine the role of the liver X receptor (LXR) in conjunction with the ABCA1 antibody, another group at the University Hospital Essen also used LXR agonists to stimulate not only ABCA1 and ABCG1, but also ABC transporters B1 (ABCB1 or MDR1) and C1 (ABCC1 or MRP1) but in a brain capillary model mimicking the blood-brain barrier (6).
