Cell cycle block under PHD3 depletion is accompanied by p27 induction. a PHD3 depletion induces a cell cycle block in G0/G1. HeLa and renal cell adenocarcinoma cells (786-O) were transfected with control (siScr) or PHD3 targeted (siPHD3) siRNA followed by synchronization at G0 and 24-h hypoxic exposure. Immunocytochemistry/Immunofluorescence: Nestin Antibody (10C2) [NB300-266] - Analysis of formalin fixed, 0.1% Triton X-100 permeabilized F9 cells (Mouse teratocarcinoma) using Nestin antibody (clone 10C2) at 1:50 dilution. The primary antibody was detected using Alexa Fluor 488 labeled donkey anti-mouse IgG (H+L) cross-adsorbed secondary antibody (green) and nuclei were counterstained with DAPI (blue). An isotype control set was inclubated with mouse IgG1 isotype control antibody and the above mentioned secondary antibody. Nestin antibody (clone 10C2) generated a specific membrane-cytoplasmic staining of Nestin while the isotype control samples did not show any staining under same assay conditions.

Flow Cytometry: Nestin Antibody (10C2) [NB300-266] - Intracellular flow cytometric staining of 1 x 10^6 CHO (A) and HEK-293 (B) cells using Nestin antibody (dark blue). Isotype control shown in orange. An antibody concentration of 1 ug/1 x 10^6 cells was used.

Flow Cytometry: Nestin Antibody (10C2) [NB300-266] - An intracellular stain was performed on SH-SY5Y cells with Nestin [10C2] Antibody NB300-266C (blue) and a matched isotype control (orange). Cells were fixed with 4% PFA and then permeabilized with 0.1% saponin. Cells were incubated in an antibody dilution of 2.5 ug/mL for 30 minutes at room temperature. Both antibodies were conjugated to DyLight 650.

Flow Cytometry: Nestin Antibody (10C2) [NB300-266] - An intracellular stain was performed on HeLa cells with Nestin Antibody (10C2)NB300-266PE (blue) and a matched isotype control (orange). Cells were fixed with 4% PFA and then permeabilized with 0.1% saponin. Cells were incubated in an antibody dilution of 2.5 ug/mL for 30 minutes at room temperature. Both antibodies were conjugated to phycoerythrin.

Nestin was detected in immersion fixed undifferentiated rat cortical stem cells using Mouse Anti-Rat Nestin Monoclonal Antibody (Catalog # MAB2736) at 10 µg/mL for 3 hours at room temperature. Cells were stained using the NorthernLights™ 557-conjugated Anti-Mouse IgG Secondary Antibody (red; Catalog # NL007) and counterstained with DAPI (blue). Zfp322a can enhance OSKM reprogramming and replace Sox2.(A) Zfp322a enhanced reprogramming efficiency and accelerated the onset of reprogramming process. OSKM serves as control experiment. (B) The iPSCs generated from OSKM plus Zfp322a presented alkaline phosphatase activity. There were more AP stained colonies generated from OKSM+Zfp322a compare to OKSM. (C) The iPSCs expressed endogenous Oct4, Nanog, Sox2, Rex1 and SSEA-1, indicating that they were ES-cell like. Immunostaining using anti-Oct4, anti-Nanog anti-Sox2, anti-Rex1 and anti-SSEA-1 antibodies were performed with GFP+ iPSCs generated from OKSM+Zfp322a. (D) GFP+ iPSCs generated by OKSM+Zfp322a were able to express ectoderm, mesoderm and endoderm lineage markers in the EB formation assay. iPSCs were stained with anti-Nestin, anti-Gata4 and anti-alpha smooth muscle actin (SMA) antibodies and pictures were taken at 60× magnification. DAPI (blue) served as nucleus marker. (E) Zfp322a was able to replace Sox2, but not Oct4 or Klf4 in OSKM reprogramming process. Results from three independent experiments were presented. Immunohistochemistry: Nestin Antibody (4D11) [NBP1-92717] - Staining in mouse cortex microslice section (from postnatal day 3). IHC image submitted by a verified customer review.

Immunocytochemistry/Immunofluorescence: Nestin Antibody (4D11) [NBP1-92717] - Mixed cultures of neonatal rat neurons and glia stained with NBP1-92717 (red), chicken antibody to Vimentin (NB300-223, green) and DNA (DAPI stain, blue). Astrocytes and neuronal stem cells stain strongly and specifically in a clearly filamentous fashion with the Nestin (4D11) antibody. The filamentous staining pattern is as expected as both Nestin and Vimentin are components of 10nm filaments. Note that some cells contain Nestin, but do not stain strongly for Vimentin and so appear red. Others contain Vimentin and not Nestin and so appear green - these are likely to be fibroblastic or endothelial cells. Some cells express both proteins and so appear yellowish.

Nestin was detected in immersion fixed rat cortical stem cells using 10 µg/mL Goat Anti-Rat Nestin Antigen Affinity-purified Polyclonal Antibody (Catalog # AF2736) for 3 hours at room temperature. Cells were stained with the NorthernLights™ 557-conjugated Anti-Goat IgG Secondary Antibody (red; Catalog # NL001) and counterstained (green). beta -III Tubulin and Nestin were detected in rat cortical stem cells (Catalog # NSC001) using 5 µg/mL neuron-specific Mouse beta -III Tubulin Monoclonal (clone TuJ-1) Antibody (Catalog # MAB1195) and 10 µg/mL Goat Anti-Rat Nestin Antigen Affinity-purified Polyclonal Antibody (Catalog # AF2736). Cells were incubated with primary antibodies for 3 hours at room temperature. Cells were stained for beta-III Tubulin using the NorthernLights™ 557-conjugated Anti-Mouse IgG Secondary Antibody (red; Catalog # NL007) and for Nestin using the NorthernLights 493-conjugated Anti-Goat IgG Secondary Antibody (green; Catalog # NL003).
