Background

SOX10 is a member of the SOX (SRY-related HMG-box) family of transcription factors involved in the regulation of embryonic development and in the determination of the cell fate. This protein may act as a transcriptional activator after forming a protein complex with other proteins. It acts as a nucleocytoplasmic shuttle protein and is important for neural crest and peripheral nervous system development. Mutations in this gene are associated with Waardenburg-Shah and Waardenburg-Hirschsprung disease.

Cells were stained using the NorthernLights™ 557-conjugated Anti-Goat IgG Secondary Antibody (red; Catalog # NL001) and counterstained with DAPI (blue). Specific staining was localized to nuclei. A172 human glioblastoma cell line were fixed using formaldehyde, resuspended in lysis buffer, and sonicated to shear chromatin. Olig2/DNA complexes were immunoprecipitated using 5 μg Goat Anti-Human/Mouse/Rat Olig2 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF2418) or control antibody (Catalog # AB-108-C) for 15 minutes in an ultrasonic bath, followed by Biotinylated Anti-Goat IgG Secondary Antibody (Catalog # BAF109). Immunocomplexes were captured using 50 μL of MagCellect Streptavidin Ferrofluid (Catalog # MAG999) and DNA was purified using chelating resin solution. The p21 promoter was detected by standard PCR.
