Background
Premelanasome protein (PMEL) is a nonmutated "self" antigen
expressed by melanocytes, most melanoma cells, substantia nigra, and retinal pigmented
epithelium. This protein plays an
essential role in the structural organization of premelanosomes. PMEL is
over-expressed in all stages of melanoma progression and is negative prognostic
marker for the overall survival rate of melanoma patients. PMEL is also associated with melanin-based
pigmentation, neurodegenerative disease (functional amyloid) and glaucoma
(missense mutations). How PMEL amyloid is tolerated within cells is of major
therapeutic interest as it is related to Alzheimer's and Parkinson's disease.
