BALB/c splenocytes were stimulated for 3 days with plate-bound Anti-Mouse CD3e Functional Grade Purified (cat. 16-0031), soluble Anti-Mouse CD28 Functional Grade Purified (cat. 16-0281), Mouse IL-2 Recombinant Protein (cat. 14-8021), and Mouse IL-4 Recombinant Protein (cat. 14-8041). Cells were then restimulated with Cell Stimulation Cocktail (plus protein transport inhibitors) for 5 hours. Following restimulation, cells were fixed and permeabilized then stained with Anti-Mouse CD4 FITC (cat. 11-0042) and 0.125 ug of Rat IgG1 K Isotype Control PE (cat. 12-4301) or 0.125 ug of Anti-Mouse IL-13 PE. | Application Notes | The eBio13A antibody has been tested as the capture antibody in a sandwich ELISA (and ELISPOT) for analysis of mouse IL-13, in combination with the biotinylated eBio1316H antibody for detection (13-7135) and recombinant mouse IL-13 (14-8131) as the standard. A suitable range of concentrations of this antibody for ELISA capture is 1.0 - 4.0 ug/ml. A standard curve consisting of doubling dilutions of the recombinant standard over the range of 1000 pg/ml - 8.0 pg/ml should be included in each ELISA plate. The Functional Grade Purified eBio1316H antibody (16-7135) is recommended for in vitro blocking studies. |
