  1. Biotinylated Protein C Antibody (100x): A 100-fold concentrated biotinylated polyclonalantibody against Protein C.
  2. Chromogen Substrate: A ready-to-use stabilized peroxidase chromogen substratetetramethylbenzidine.
  3. Human Protein C Microplate: A 96-well polystyrene microplate (12 strips of 8 wells) coatedwith a polyclonal antibody against human Protein C.
  4. Human Protein C Standard: Human Protein C in a buffered protein base (lyophilized).
  5. Sealing Tapes: Precut, pressure-sensitive sealing tapes that can be cut to fit the format ofthe individual assay.
  6. Stop Solution: A 0.5 N hydrochloric acid to stop the chromogen substrate reaction.
  7. Streptavidin-Peroxidase Conjugate (SP Conjugate): A 100-fold concentrate.
  8. Wash Buffer Concentrate (20x): A 20-fold concentrated buffered surfactant.

Background

Protein C is a vitamin K-dependent serine protease that is produced in the liver and made up of 2 polypeptide chains. The 62 kDa proenzyme is activated by thrombin bound to an endothelial surface receptor and the active enzyme cleaves factor Va and VIIIa inhibiting blood coagulation. The molecular weight of the active enzyme is 55 kDa and the normal concentration in human plasma is approximately 1-3 ng/ml while the concentration of the proenzyme is approximately 3 ug/mL due to the very fast turnover.
