![Block/Neutralize: TLR4 Antibody (HTA125) [NB100-56723] - Validation of the neutralizing effect of this antibody on TLR4 activity. The TLR4/MD-2/CD14/IL-8 Prom/LUCPorter HeLa cells were plated in 96-well white plates at 1 x 10^5 cells/well. After 16 h, cells were preincubated with different amounts of this antibody for 1 h and then stimulated with 10 ng/ml LPS for 6 h. Luciferase activity was then directly analyzed using the one-step luciferase reporter assay reagent. Data Summary: It inhibited LPS-mediated TLR4 activity in the TLR4/MD-2/CD14/IL-8 Prom/LUCPorter cell line. Negative (LPS-, LPS-/CP7+, LPS-/CP7+) and positive (LPS+) controls were also included. Supernatants were harvested after 6 h and murine TNF-alpha measured by ELISA. ![Immunocytochemistry/Immunofluorescence: TLR4 Antibody [NBP2-24821] - U2OS cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X TBS + 0.5% Triton X-100. The cells were incubated with anti-TLR4 [NBP2-24821] at a 1:50 dilution overnight at 4C and detected with an anti-rabbit DyLight 488 (Green) at a 1:500 dilution. Alpha tubulin (DM1A) NB100-690 was used as a co-stain at a 1:1000 dilution and detected with an anti-mouse DyLight 550 (Red) at a 1:500 dilution. Nuclei were counterstained with DAPI (Blue).
