Background

In contrast to fast and slow skeletal muscle cells that fuse and terminally differentiate, smooth muscle cells are able to simultaneously proliferate and express lineage-restricted proteins. One of these proteins, expressed exclusively in smooth muscles, has been referred to as SM22-alpha, a 22 kDa protein with structural similarity to the vertebrate thin filament myofibrillar regulatory protein calponin and the Drosophila muscle protein mp20, neither of which play a direct role in the contractile apparatus.The protein is a transformation and shape-change sensitive actin cross-linking/geling protein found in fibroblasts and smooth muscle. Its expression is down-regulated in many cell lines, and this down-regulation may be an early and sensitive marker for the onset of transformation. A functional role of this protein is unclear.

  1. We are using the Transgelin antibody (NB600-507) for western analysis of mouse and human uterine tissue. In the mouse we see a single band of 22 kDa but in the human we see a band of 12 kDa. Any thoughts on this? I can not find any information on a smaller transgelin band.
    • I have searched for an explanation for the lower molecular weight band you are detecting, and have found that this paper, PMC2789179 says: 'It should be noted that the expression analysis of TAGLN is complicated by the fact that multiple bands were detected on the Western blots. Similar observations have been described and attributed to C-terminal proteolysis. While the sum of band intensities was used for the analyses here, it is possible that TAGLN regulation is not a simple change in quantity of one defined species, but may, in fact, be subject to posttranslationally regulated events such as modification or degradation.'
