  1. I received an inquiry about ChromataChIP Kit. Regarding phase 3(p.6), what size tube should be used? And what size tubes can be used for Magnetic tube separator rack (Cat# NBP1-72032)?
    • Generally this step is performed using 1.5mL tubes. The stand can hold eight 1.5-ml tubes or one 15-ml tube, and one 50-ml tube.
  2. I have one question about ChIP. In your Chromata ChIP kit, you add 25ul magnetic beads to the samples. Is there any way to normalize for differences in the amounts of magnetic beads in each sample? For example, it can happen that one sample got a bit more beads than another, is there some way to take this into account when you analyze your data?
    • As long as the vial/bottle of beads is vortexed so that the beads are fully resuspended in solution before taking the 25ul out, there will not be any significant variation in bead number. However, if you are taking multiple 25ul aliquots out, it is best to quickly revortex before every aliquot pipetting. For all of our internal testing, we typically see a CV of less than 2% for both intra- and interassay variation, which is very good.
  3. I am planning ChIP experiments using a rat cell line and also rat tissues. I am interested in your Chromata ChIP kit but I have some questions. The magnetic tube separator rack: is it possible to use a magnetic separator from Miltenyi Biotech (we already have one)? Is the positive control antibody (H3K4Me3) and primers (human RPL30) would work with rat samples? If no, could you provide a positive control working with the rat specie or give the primers sequences? Finally, what are the changes that have to be made to the protocol for ChIP assays starting from tissues? (Only an example using cells is given in the protocol)
    • 1. Yes, your magnetic rack will work just fine. 2. Unfortunately, our primers do not work with species other than human and we do not have any other primers available. You should be able to design your own to the same target in rat though and it should be just fine. 3. We are in the process of launching tissue specific protocols for use in ChIP. These are not available online yet but if you contact our Technical Team we can assist you with this
  4. Regarding the ChromataChIP Kit cat.#NBP1-71709: 1. What antibodies do you have? 2. Do you supply the qPCR primers for humans (positive and negative targets for each antibody)? 3. Do you supply the negative antibody? I like your guarantee, but hope never to hold you to it!
    • I am glad you are interested in our ChromataChIP Kits and would be happy to answer your questions. We have several ChIP grade epigenetics antibodies available and this link should take you to additional information about the kit itself and related products. I don't know if you saw this page first before inquiring further about the kit. Let me know if so and you still had additional questions and I would be happy to address those with the lab person who developed the kit. We are proud of our guarantee and of course prefer that no one would have to use it, but sometimes that is inevitable and as long as we can resolve any issues that arise are happy to do solid completely understand and know that is where your focus and attention will be. Let us know once you have more time on your hands if you are still interested in the lab visit and I can coordinate that with the lab.
  5. I wanted to know whether the H3K4me3 positive control antibody has cross reactivity with other species? I am using mouse tissue and would like to know whether this antibody will still work?
    • The positive control antibody in this kit is expected to cross-react with mouse, rat, c. elegans and most other mammals. It will work just fine for mouse.
  6. Do you know of anyone that has used this kit for plant tissues, preferably intact tissues? Also, do you have references where people have used your ChIP kit available?
    • We don't know of anyone using this kit on plants at this time. H3 is very well conserved, even in plants, as the immunogen to generate the antibody shares 100% similarity with tobacco, rice, maize and most other plants. This kit will almost certainly work with your plant. The main problem is that the primers almost certainly won't work on plants. Even one base pair difference can prevent the primers from working. Chances are there is some dissimilarity at the DNA level that doesn't effect the protein sequence because of the wobble rule. You can most likely use this kit on your plant, but you will need to design your own control primers.
