Mapk Cascade Pathway Bioinformatics

The MAPK pathway is made up of multiple cascades which can lead to a variety of cellular functions including proliferation, differentiation, apoptosis, and cell survival. There are a number of different stimuli and factors that can initiate the MAPK cascades, such as growth factors that bind to RTKs, proinflammatory cytokines, heat shock, and osmotic stress. The signaling molecule causes dimerization of the RTKs, which causes the binding of GRB2 and SOS leading to the Ras-GDP complex to become Ras-GTP. This initiates a phosphorylation cascade of molecules such as MEK and ERK, which activate transcription factors in the AP1 family and begin the process of growth and proliferation. In typical instances of the MAPK pathway, the Ras-GTP complex is inactivated shortly after its activation. When this process is deregulated, it can lead to the increase in the proliferation and cell survival that is seen in progressing tumors.
