Hoist the DYKDDDDK Epitope Tag

Thu, 09/05/2013 - 14:50

The DYKDDDDK Epitope Tag antibody was raised against a short, artificial sequence that is widely and commonly used as a protein fusion tag. It is pervasively present in a vast amount of scientific studies, and is a very powerful investigative research tool for applications such as immunofluorescence, immunoprecipitation, Western blot, and immunohistochemistry. The DYKDDDDK Epitope Tag antibody was engineered by Park's group at the Rockefeller University as a more superior antibody for improved detection and sensitivity in a wide variety of methods and investigative analyses (1). This antibody was assessed and validated for efficacy in various immunodetection methods, especially immunofluoresence immmunohistochemistry and flow cytometry.

Hubbi, et. al. used the DYKDDDDK Epitope Tag antibody to dissect the interacting domains required for HIF-1 transactivation and were able to establish a direct link between the MCM proteins and cell-cycle machinery (2). In Kuver’s lab, DYKDDDDK Epitope Tag antibody was used to measure expression of a tagged construct involving the alpha4beta2gamma receptors (3). They were able to closely follow the trafficking of surface expression of these receptors in their high efficacy states in cultured hippocampal neurons. Idoyaga’s group used the DYKDDDDK Epitope Tag antibody to engineer a new hybrid antibody that targets the extracellular domain of mouse Langerin receptor (4). The hybrid antibody was used to evaluate Langerin function in dendritic cells (DCs) in dose response studies.
