CRLF2 - a receptor for thymic stromal lymphopoietin (TSLP)

CRLF2 has been reported as an important factor that drives dendritic cell maturation and activation. It was originally shown to participate in the positive selection of regulatory T-cells, maintenance of peripheral CD4+ T-cell homeostasis, and induction of CD4+ T cell-mediated allergic reactions. CRLF2 binds to its receptor with low-affinity; high-affinity binding is facilitated by the presence of IL-7R-alpha to form a functional heteromeric complex. This protein is also capable of supporting the growth of fetal liver and adult B-cell progenitors. CRLF2 is now known to have wide-ranging roles not just in these hematopoietic cell lineages, but also in multiple disease states within multiple organ systems.

Recent advances highlighting the newly attributed and unexpected regulatory role of CRLF2 in a variety of cancers, both solid and leukemia, are shared in a new review from Lo Kuan’s lab at the Benaroya Research Institute (1). Additionally, the same research group published a review that compiles the most up-to-date knowledge of CRLF2 signal transduction and how it impacts conditions such as allergies (lung- and skin-specific subtypes) and autoimmunity (2).  Morshed et al from the Bern University Hospital performed immunoblot and immunohistochemical studies with the CRLF2 antibody in their analyses of eosinophil extracellular trap (EET) release (3). Using the CRLF2 antibody, they were able to demonstrate that CRLF2 directly stimulates EETs in both peripheral blood and tissue infiltrating eosinophils, and that this is a key component of antibacterial activity in skin disease. This data is helpful in linking environmentally-triggered CRLF2 expression to pathogen defense mechanisms found in eosinophils.
