Application Notes
In A375, a ~50 kDa band is observed. The CCR10 antibody is made against an epitope in the cytoplasmic domain of the receptor (Moed et al, 2004). For flow cytometry (intracellular) Moed et al fixed cells with 2% formaldehye and permeabilized with 0.1% saponin.
In Simple Western only 10 - 15 uL of the recommended dilution is used per data point. Separated by Size-Wes, Sally Sue/Peggy Sue.
Store at 4C short term. Aliquot and store at -20C long term. Avoid freeze-thaw cycles.

Background
Recently, a function has been assigned to a previously identified orphan chemokine receptor, GPR-2. This receptor, also called CCR10, binds with monocyte chemoattractant protein (MCP)-1, MCP-3 and MIP-1 beta. CCR10 is expressed by melanocytes, dermal fibroblasts, dermal microvascular endothelial cells, T cells as well as in skin-derived Langerhans cells. CCR10 may play a role in both skin homeostasis as well as a potential role in inflammatory responses.
